Description
Citicoline is a nucleotide joined to choline through a pyrophosphate bridge, and almost every practical difficulty with the material follows from that bridge. The table sets each catalogue parameter against the consequence it carries at the bench.

The pyrophosphate linkage is the weak point of the molecule, and the two markers of its cleavage are the corresponding nucleoside monophosphate and phosphocholine. A separation that cannot resolve those two from the parent compound is not a stability-indicating one, however clean the chromatogram looks. Free choline is worth determining separately, because it responds poorly in ultraviolet and can hide entirely behind the strong absorption of the cytosine ring. Phosphorylated nucleotide materials of this kind are commonly hygroscopic, so water content and the handling conditions named on the certificate deserve more attention than the assay figure. Weigh quickly, close the container between operations and keep the reserve sealed. Solutions prepared in water should be dated and stored cold, since hydrolysis at the bridge continues quietly at room temperature and shows up first as a small early-eluting peak rather than as a change in appearance.
