Confirming the labdane skeleton
The molecule is a highly oxygenated bicyclic diterpene: several hydroxyl groups, one acetate ester and a vinyl substituent on a rigid decalin-derived framework. Identity is confirmed against a certified reference standard rather than assumed from a label. Three independent lines normally close the question — retention agreement in a reversed-phase system, an infrared spectrum superimposable with the standard, and resonance patterns consistent with the polyoxygenated framework. Where no standard is on hand, the container stays marked as identity-pending in the register.
Assay and chromatographic separation
This diterpene carries no strong chromophore, so ultraviolet detection works only at short wavelengths, where the mobile phase itself starts to absorb. Control laboratories therefore run charged aerosol or evaporative light scattering detection alongside low-wavelength ultraviolet, using an acetonitrile-water gradient on an octadecyl phase. Area normalisation returns a chromatographic purity figure, which is not the same quantity as an assay against a standard, and the certificate has to say which of the two it quotes. Impurities carried over from the plant matrix
Material of botanical origin brings a family of structurally close congeners with it — isoforskolin, the deoxy analogues and related labdanes formed alongside the main compound in the root. They elute near the principal peak and are the usual reason a batch misses a tight specification. Beyond them, a complete batch file covers residual solvents from extraction and crystallisation, water content by Karl Fischer titration, sulphated ash and elemental impurities. Chromatographic purity on its own therefore never closes the assessment.
- Let the sealed pouch reach ambient temperature before opening, so that moisture does not condense on cold solid.
- Weigh in a low-humidity area with an anti-static spatula, because fine crystalline powder creeps across balance pans.
- Prepare stock solutions in an aprotic solvent, in amber glass, and record the exact mass and volume used.
- Split the gram into working aliquots so the main container is opened once rather than twenty times.
- Log every withdrawal with date, mass, operator and the experiment it feeds.
Batch documentation and traceability
A usable certificate names the batch, the packaging date, the retest date, the technique behind every figure and the person who released the lot.
